SP8-STED: Leica TCS SP8 laser scanning confocal microscope with STED super-resolution

Laser scanning confocal microscope with spectral emission detection (Leica TCS SP8) on an inverted stand, fully motorized stage, fast z movement (Leica Super Z Galvo stage). Resonant scanner (8 Hz) for fast scan times and lower photo-toxicity or bleaching. White light laser allows free choice of excitation wavelength between 470 and 670 nm. STED (stimulated emission-depletion) super-resolution through two depletion lasers, 592 and 775 nm. Five internal detectors (two HyD high-sensitivity with time gating, three PMT), one transmitted light detector (PMT). Environmental control of temperature, CO2, humidity, possible through a mobile stage-top incubator (Tokai Hit).

Lasers for fluorescence excitation:

405 nm for blue emitters
442 nm for cyan emitters
470-670 nm pulsed white light laser for fluorophores from blue-green to far-red emission

Lasers for STED depletion:

592 nm continuous wave (non-pulsed) laser
775 nm pulsed laser
